Yesterday, I was noticed that the vinculin expression constructs are transformed into the DH5alpha, not BL21. And I found that the purified protein degradated as predicted. Overnight exposure did not get any bands. So, maybe I will start a new purification cycle next week.
One thing is good, that I have transformed the coronin expression vector into BL21 CodonPlus RP. Hope I can get some good information.
